Association of apolipoprotein E polymorphism with plasma lipids and Alzheimer's disease in a Southern Brazilian population BJMBR
de-Andrade,F.M.; Larrandaburu,M.; Callegari-Jacques,S.M.; Gastaldo,G.; Hutz,M.H..
Apolipoprotein E (protein: apo E; gene: APOE) plays an important role in the multifactorial etiology of both Alzheimer's disease (AD) and lipid level concentrations. The polymerase chain reaction (PCR) was used to investigate the APOE gene polymorphism in 446 unrelated Caucasians, among them 23 AD patients, and 100 Afro-Brazilians living in Porto Alegre, Brazil. The frequencies of the APOE*2, APOE*3 and APOE*4 alleles were 0.075, 0.810 and 0.115 in Caucasians and 0.075, 0.700 and 0.225 in Afro-Brazilians, respectively (c2 = 8.72, P = 0.013). A highly significant association was observed between the APOE*4 allele and AD in this population-based sample. The APOE*4 frequency in AD patients (39%) was about four times higher than in the general Caucasian...

Effect of neoadjuvant chemotherapy on low-density lipoprotein (LDL) receptor and LDL receptor-related protein 1 (LRP-1) receptor in locally advanced breast cancer BJMBR
Pires,L.A.; Hegg,R.; Freitas,F.R.; Tavares,E.R.; Almeida,C.P.; Baracat,E.C.; Maranhão,R.C..
Low-density lipoprotein (LDL) receptors are overexpressed in most neoplastic cell lines and provide a mechanism for the internalization and concentration of drug-laden nanoemulsions that bind to these receptors. The aim of the present study was to determine whether the administration of standard chemotherapeutic schemes can alter the expression of LDL and LDL receptor-related protein 1 (LRP-1) receptors in breast carcinoma. Fragments of tumoral and normal breast tissue from 16 consecutive volunteer women with breast cancer in stage II or III were obtained from biopsies before the beginning of neoadjuvant chemotherapy and after chemotherapy, from fragments excised during mastectomy. Tissues were analyzed by immunohistochemistry for both receptors. Because...

Effects of rosiglitazone on serum paraoxonase activity and metabolic parameters in patients with type 2 diabetes mellitus BJMBR
Atamer,Y.; Atamer,A.; Can,A.S.; Hekimoğlu,A.; Ilhan,N.; Yenice,N.; Koçyiğit,Y..
Human serum paraoxonase contributes to the anti-atherogenic effect of high-density lipoprotein cholesterol (HDL-C) and has been shown to protect both low-density lipoprotein cholesterol (LDL-C) and HDL-C against lipid peroxidation. We investigated the effects of rosiglitazone on paraoxonase activity and metabolic parameters in patients with type 2 diabetes mellitus [50 patients (30 males, 20 females); mean±SD age: 58.7±9.2 years, body mass index: 28.2±4.1'kg/m2], in whom glucose control could not be achieved despite treatment with metformin, sulphonylurea, and/or insulin. The patients were given 4'mg/day rosiglitazone for 3 months in addition to their usual treatment. Serum paraoxonase activity, malondialdehyde, homocysteine, and lipid profile were...

Effet du taux et de la nature des lipides alimentaires sur les mécanismes intervenant dans la constitution des dépôts lipidiques (transport, captage, synthèse) chez la truite arc-en-ciel et le bar ArchiMer
Richard, Nadège.
Replacement of dietary fish oil by a blend of vegetable oils for more than a year does not affect growth, hepatic lipogenesis and tissue lipid uptake in rainbow trout and European seabass. However, this replacement decreases plasma cholesterol level in both species and LDL amount in rainbow trout. Such replacement does not affect muscle lipid content of fish but modifies fatty acid composition of fish flesh which reflects in most part that of dietary lipid source. Increasing dietary lipid level inhibits hepatic lipogenesis but does not modify tissue lipid uptake in seabass and in two trout lines selected for muscle lipid level. In seabass, dietary lipid level had a lower impact on growth and lipid metabolism than rearing temperature. In this species,...

Higher values of triglycerides:HDL-cholesterol ratio hallmark disease severity in children and adolescents with sickle cell anemia BJMBR
Teixeira,R.S.; Arriaga,M.B.; Terse-Ramos,R.; Ferreira,T.A.; Machado,V.R.; Rissatto-Lago,M.R.; Silveira-Mattos,P.S.; Boa-Sorte,N.; Ladeia,A.M.T.; Andrade,B.B..
Dyslipidemia has been described in sickle cell anemia (SCA) but its association with increased disease severity is unknown. Here, we examined 55 children and adolescents with SCA as well as 41 healthy controls to test the association between the lipid profiles in peripheral blood and markers of hemolysis, inflammation, endothelial function, and SCA-related clinical outcomes. SCA patients exhibited lower levels of total cholesterol (P<0.001), low-density lipoprotein cholesterol (LDL-c) (P<0.001), and high-density lipoprotein cholesterol (HDL-c) (P<0.001), while displaying higher triglyceride (TG) levels and TG/HDL-c ratio values (P<0.001). TG/HDL-c values were positively correlated with lactate dehydrogenase (P=0.047), leukocyte count (P=0.006),...

L'absorption intestinale des lipides chez la truite arc-en-ciel (Salmo gairdneri ) ArchiMer
Vernier, J; Sire, M.
Though the absorption of dietary lipids is much slower in S. gairdneri than in mammals, the mechanism does not differ fundamentally. The long chain fatty acids resulting from intraluminal hydrolysis are esterified by the epithelial cells and transferred as VLDL-like particles and chylomicrons to lymph. The size of granules depends on the nature of the ingested fatty acids. The intracellular stages of this lipoprotein synthesis are defined.

Lipolysis of emulsion models of triglyceride-rich lipoproteins is altered in male patients with abdominal aorta aneurysm BJMBR
Hosni,J.J.; Vinagre,C.G.; Mady,C.; Maranhão,R.C..
Disorders of the lipid metabolism may play a role in the genesis of abdominal aorta aneurysm. The present study examined the intravascular catabolism of chylomicrons, the lipoproteins that carry the dietary lipids absorbed by the intestine in the circulation in patients with abdominal aorta aneurysm. Thirteen male patients (72 ± 5 years) with abdominal aorta aneurysm with normal plasma lipid profile and 13 healthy male control subjects (73 ± 5 years) participated in the study. The method of chylomicron-like emulsions was used to evaluate this metabolism. The emulsion labeled with 14C-cholesteryl oleate and ³H-triolein was injected intravenously in both groups. Blood samples were taken at regular intervals over 60 min to determine the decay curves. The...

Peripheral brain-derived neurotrophic factor is related to cardiovascular risk factors in active and inactive elderly men BJMBR
Zembron-Lacny,A.; Dziubek,W.; Rynkiewicz,M.; Morawin,B.; Woźniewski,M..
Regular exercise plays an important preventive and therapeutic role in heart and vascular diseases, and beneficially affects brain function. In blood, the effects of exercise appear to be very complex and could include protection of vascular endothelial cells via neurotrophic factors and decreased oxidative stress. The purpose of this study was to identify the age-related changes in peripheral brain-derived neurotrophic factor (BDNF) and its relationship to oxidative damage and conventional cardiovascular disease (CVD) biomarkers, such as atherogenic index, C-reactive protein (hsCRP) and oxidized LDL (oxLDL), in active and inactive men. Seventeen elderly males (61-80 years) and 17 young males (20-24 years) participated in this study. According to the 6-min...

Prostaglandin F2α Increases Lipoprotein Utilization for Progesterone in Bovine Early Corpora Lutea In Vitro OAK
KOBAYASHI, Shu-ichi; MIYAMOTO, Akio; 宮本, 明夫.
Luteal cells utilize lipoproteins (LIPs) for progesterone (P) synthesis. A luteolysin prostaglandin F2α (PGF 2α) has shown to prevent the LIPs utilization for steroidogenesis in bovine midcycle luteal cells. However, a luteolytic injection of PGF2α before Day 5 (Day 0=estrus) can not induce the regression of the corpus luteum (CL). This suggests that the intraluteal mechanism of PGF2 α action on the LIPs utilization in early CL is different from that of the mid luteal stage. Thus, this study aimed to investigate the interaction between PGF2 α and LIPs on the secretion of P and oxytocin (OT) from bovine early CL. An in vitro microdialysis system (MDS) was used as a model. Each bovine early CL (Days 3-4) was implanted with the MDS, and maintained in an organ...

Reversible flow of cholesteryl ester between high-density lipoproteins and triacylglycerol-rich particles is modulated by the fatty acid composition and concentration of triacylglycerols BJMBR
Cazita,P.M.; Castilho,L.N.; Carvalho,M.D.T.; Sesso,A.C.; Oliveira,H.C.F.; Quintão,E.C.R..
We determined the influence of fasting (FAST) and feeding (FED) on cholesteryl ester (CE) flow between high-density lipoproteins (HDL) and plasma apoB-lipoprotein and triacylglycerol (TG)-rich emulsions (EM) prepared with TG-fatty acids (FAs). TG-FAs of varying chain lengths and degrees of unsaturation were tested in the presence of a plasma fraction at d > 1.21 g/mL as the source of CE transfer protein. The transfer of CE from HDL to FED was greater than to FAST TG-rich acceptor lipoproteins, 18% and 14%, respectively. However, percent CE transfer from HDL to apoB-containing lipoproteins was similar for FED and FAST HDL. The CE transfer from HDL to EM depended on the EM TG-FA chain length. Furthermore, the chain length of the monounsaturated...

The beta-chemokines MIP-1alpha and RANTES and lipoprotein metabolism in HIV-infected brazilian patients BJID
Mikawa,Angela Yumico; Malavazi,Iran; Tagliavini,Sandra Antonia; Abrão,Emiliana P.; Costa,Paulo Inácio da.
HIV patients are predisposed to the development of hypertriglyceridemia and hypercholesterolemia as a result of both viral infection and HIV infection therapy, especially the protease inhibitors. Chemokines and cytokines are present at sites of inflammation and can influence the nature of the inflammatory response in atherosclerosis. We investigated the correlation between biochemical variables and beta-chemokines (MIP-1alpha and RANTES) and the apolipoprotein E genotype in HIV-infected individuals. The apolipoproteins were measured by nephelometry. Triglycerides and total cholesterol were determined by standard enzymatic procedures. The beta-chemokines were detected by ELISA. The genetic category of CCR5 and apolipoprotein E were determined by PCR...
